A quest to discover new anaerobic microbes (and their lipids !) in the Black Sea

The Black Sea is the largest permanently stratified euxinic basin in the world, characterized by the presence of multiple redox gradients, which makes it an excellent setting to detect anaerobic microbes with novel metabolic strategies. We are focused on detecting unknown yet-uncultured anaerobic microorganisms by using ‘omics’ approaches such as metagenomics and lipidomics, as well as incubations with labeled substrates combined with stable isotope probing. Moreover, we are aiming to discover new membrane lipids of uncultured anaerobic microbes that could be used as biomarkers for their presence, both in present and past microbial communities.

Here, I will introduce the challenges of seagoing expeditions to the Black Sea and the data derived from our sampling campaigns. By applying ‘omic’ methods we have detected a high diversity of bacteria and archaea involved in organic matter processing and sulfur cycling which will guide future culturing efforts.
